About

Afatinib, often branded as Afzeta, is an irreversible ErbB family blocker, categorized as a second-generation tyrosine kinase inhibitor (TKI). It works by irreversibly binding to and inhibiting the signaling pathways mediated by epidermal growth factor receptor (EGFR), HER2, and HER4. This comprehensive inhibition effectively blocks cell proliferation, survival, and angiogenesis, while promoting apoptosis in cancer cells that overexpress these receptors. Afatinib is primarily indicated for the treatment of specific types of non-small cell lung cancer (NSCLC) that harbor common activating EGFR mutations, such as exon 19 deletions or L858R substitutions. Its targeted action provides a significant therapeutic advantage by directly addressing the molecular drivers of the cancer, leading to improved progression-free survival and overall response rates in carefully selected patient populations. It represents a cornerstone in personalized oncology for EGFR-mutated NSCLC.
