About
Acyclovir is a highly effective antiviral medication primarily indicated for the treatment of infections caused by the herpes simplex virus (HSV) and varicella-zoster virus (VZV). This synthetic purine nucleoside analogue exerts its therapeutic effect by selectively inhibiting viral DNA replication. Upon entering a virally infected cell, acyclovir is converted to its active triphosphate form by viral thymidine kinase, an enzyme predominantly found in herpesvirus-infected cells. The active triphosphate then competitively inhibits viral DNA polymerase and is incorporated into the nascent viral DNA chain, leading to premature chain termination and irreversible inactivation of the viral enzyme. This selective activation mechanism ensures potent antiviral activity against herpesviruses while minimizing toxicity to uninfected host cells. Acyclovir helps to significantly reduce the severity, duration, and frequency of herpes outbreaks, and is also valuable for prophylactic use in immunocompromised individuals to prevent recurrent infections.
