About
Temozolomide, known as Temorel, is an oral alkylating agent used in the treatment of certain brain tumors. It is a prodrug that undergoes rapid chemical conversion at physiological pH to its active metabolite, monomethyl triazenoimidazole carboxamide (MTIC). MTIC then methylates DNA at various sites, primarily at the O6 and N7 positions of guanine. This methylation leads to DNA damage and subsequent apoptosis (programmed cell death) in rapidly dividing cancer cells. Its ability to cross the blood-brain barrier makes it particularly effective against primary and metastatic brain tumors.
Temorel is a crucial component of chemotherapy regimens for aggressive cancers such as glioblastoma multiforme and anaplastic astrocytoma, improving progression-free survival and overall survival in these challenging malignancies. It is often used in combination with radiotherapy for newly diagnosed glioblastoma. The oral administration offers convenience for patients, while its targeted DNA damaging effect provides a potent therapeutic strategy against brain tumors.
