About

Mycophenolate mofetil is a prodrug that is rapidly hydrolyzed to its active metabolite, mycophenolic acid (MPA), a potent and selective immunosuppressant. It is widely used in combination with other agents, such as corticosteroids and cyclosporine or tacrolimus, to prevent acute organ rejection in patients undergoing kidney, heart, or liver transplantation. MPA exerts its immunosuppressive effects by reversibly inhibiting inosine monophosphate dehydrogenase (IMPDH), a key enzyme in the de novo pathway of guanosine nucleotide synthesis. Lymphocytes, particularly T and B cells, are critically dependent on this pathway for their proliferation and function. By depleting guanosine nucleotides, mycophenolate mofetil effectively suppresses lymphocyte proliferation, antibody production, and cell-mediated immune responses, thereby helping the body accept the transplanted organ.

Directions For Use

  • Breastfeeding - Mycophenolate mofetil is excreted into breast milk and can cause serious adverse reactions in a nursing infant. Breastfeeding is contraindicated.
  • Liver - Use with caution in patients with severe liver impairment. Dose adjustments may be necessary, and liver function should be monitored.
  • Kidney - Use with caution in patients with severe chronic kidney impairment. Dose adjustments may be required, and renal function should be closely monitored.
  • Lung - Rarely, mycophenolate mofetil has been associated with pulmonary fibrosis or interstitial lung disease. Patients should report any new or worsening respiratory symptoms to their doctor.
