About
Vinblastine is an antineoplastic (anticancer) drug that is effective as a single agent, but its therapeutic effect is enhanced when used in combination with other anticancer medications.
It is commonly used in combination therapy for Hodgkin’s disease (Stages III and IV), often with adriamycin (doxorubicin), bleomycin, and dacarbazine (ABVD regimen). Vinblastine is also used in combination with cisplatin and bleomycin for the treatment of advanced testicular carcinoma.
In addition, Vinblastine has been employed for the palliative treatment of conditions such as lymphocytic lymphoma, histiocytic lymphoma, advanced mycosis fungoides, Kaposi's sarcoma, and Histiocytosis X. It may also be used in choriocarcinoma resistant to other chemotherapy, as well as breast carcinoma unresponsive to endocrine surgery or hormonal therapy.
Vinblastine is considered one of the most effective single agents for treating Hodgkin’s disease. For previously untreated patients with advanced Hodgkin’s disease, protocols substituting cyclophosphamide for nitrogen mustard and vinblastine for vincristine in the MOPP regimen provide an alternative therapy. Patients with relapsed disease have also responded well to combination therapy that includes vinblastine.
Although advanced testicular germ-cell cancers respond to vinblastine alone, concurrent use with other anticancer agents produces superior results. The effectiveness of bleomycin is enhanced when vinblastine is administered 6–8 hours prior, as this timing allows more cancer cells to be arrested in metaphase, the phase in which bleomycin is most active.
