Xigduo XR 5/1000 is a combination medication containing dapagliflozin, a sodium-glucose co-transporter 2 (SGLT2) inhibitor, and metformin hydrochloride extended-release, a biguanide. Dapagliflozin works by increasing the excretion of glucose in the urine, thereby lowering blood glucose levels independently of insulin. Metformin primarily reduces glucose production by the liver, decreases glucose absorption from the intestine, and improves insulin sensitivity. This dual mechanism provides comprehensive glycemic control for adults with type 2 diabetes mellitus. Xigduo XR also offers significant cardiovascular and renal benefits, including a reduction in the risk of cardiovascular death, hospitalization for heart failure, and progression of kidney disease in patients with type 2 diabetes and established cardiovascular disease or chronic kidney disease. The extended-release formulation of metformin allows for once-daily dosing.

Directions For Use

  • Breastfeeding - Not recommended during breastfeeding as both components may pass into breast milk; consult a doctor.
  • Liver - Use with caution in patients with liver impairment; metformin is contraindicated in severe liver disease due to increased risk of lactic acidosis.
  • Kidney - Metformin is contraindicated in severe kidney impairment (eGFR <30 mL/min/1.73m²); dapagliflozin is not recommended below this eGFR for glycemic control.
