About

Akynzeo Capsule is a fixed-dose combination medication containing netupitant and palonosetron, specifically designed for the prevention of acute and delayed nausea and vomiting associated with highly emetogenic cancer chemotherapy (HEC) and moderately emetogenic cancer chemotherapy (MEC). Netupitant is a neurokinin-1 (NK1) receptor antagonist, which blocks the action of substance P, a key neurotransmitter involved in chemotherapy-induced nausea and vomiting (CINV). Palonosetron is a serotonin-3 (5-HT3) receptor antagonist, which blocks serotonin at peripheral vagal nerve terminals and centrally in the chemoreceptor trigger zone.

By targeting two distinct pathways involved in CINV, Akynzeo provides comprehensive and prolonged antiemetic protection. Its single-dose regimen per chemotherapy cycle simplifies administration and enhances patient adherence, significantly improving the quality of life for cancer patients undergoing chemotherapy. It is crucial to administer Akynzeo prior to the start of chemotherapy to achieve optimal prophylactic effect.

Safety Measures

  • Alcohol - No specific interaction is known between Akynzeo and alcohol. However, alcohol can exacerbate nausea and vomiting, so it's generally advisable to avoid it during chemotherapy treatment.
  • Pregnancy - Akynzeo is not recommended during pregnancy unless the potential benefits outweigh the potential risks to the fetus. Consult your doctor, as animal studies have shown some adverse effects.
  • Breastfeeding - It is unknown if Akynzeo passes into breast milk. Due to the potential for serious adverse reactions in breastfed infants, breastfeeding is not recommended during treatment and for at least one month after the last dose.
  • Liver - No dose adjustment is required for mild to moderate hepatic impairment. However, caution is advised in severe hepatic impairment, and close monitoring for adverse effects is recommended.
  • Kidney - No dose adjustment is required for mild to moderate renal impairment. For severe renal impairment or end-stage renal disease, caution is advised, but no specific dose recommendations exist.
  • Lung - Akynzeo generally does not have direct adverse effects on lung function. However, any severe allergic reaction could potentially impact respiratory status; seek immediate medical attention if breathing difficulties occur.
