About
Rosuvastatin is a highly efficacious HMG-CoA reductase inhibitor, commonly known as a statin, primarily indicated for the management of dyslipidemia and the reduction of cardiovascular disease risk. Its mechanism involves competitively inhibiting HMG-CoA reductase, a rate-limiting enzyme in the hepatic biosynthesis of cholesterol. This inhibition leads to a significant decrease in intracellular cholesterol production, which in turn upregulates the expression of low-density lipoprotein (LDL) receptors on the surface of liver cells. The increased number of LDL receptors enhances the uptake and catabolism of circulating LDL-C, effectively lowering plasma LDL-C, total cholesterol, and triglyceride levels, while also modestly increasing high-density lipoprotein cholesterol (HDL-C). Rosuvastatin is prescribed for primary hypercholesterolemia, mixed dyslipidemia, and crucially, for the primary and secondary prevention of major cardiovascular events, including myocardial infarction and stroke, in individuals identified as being at high cardiovascular risk.
Uses
- Treatment of primary hypercholesterolemia.
- Management of mixed dyslipidemia.
- Reduction of elevated triglyceride levels.
- Prevention of major cardiovascular events in high-risk patients.

- Increased HDL-C ("good" cholesterol).
- Lowered triglyceride levels.
- Reduced risk of heart attack and stroke.
- Effective in primary and secondary prevention.
- Well-tolerated by most patients.
Side Effects

- Elevated liver enzymes (transaminases)
- Proteinuria (protein in urine)
- Rash
- Pancreatitis (rare)
- Rhabdomyolysis (rare but serious muscle breakdown)
Safety Measures
- Alcohol - Excessive alcohol consumption can increase the risk of liver problems associated with statins. Use with caution.
- Pregnancy - Contraindicated during pregnancy due to potential harm to the fetus. Women of childbearing potential should use effective contraception.
- Breastfeeding - Contraindicated during breastfeeding as it may pass into breast milk and cause adverse effects in the infant.
- Liver - Contraindicated in patients with active liver disease or unexplained persistent elevations of serum transaminases.
- Kidney - Dosage adjustment is recommended in patients with severe renal impairment (CrCl < 30 mL/min).
- Lung - Generally safe for use in patients with lung conditions, but interstitial lung disease has been reported rarely.
