About

Baclofen is a centrally acting skeletal muscle relaxant and antispastic agent, primarily used to alleviate muscle spasticity resulting from conditions such as multiple sclerosis, spinal cord injuries, and other neurological disorders. Its mechanism of action involves mimicking the effects of gamma-aminobutyric acid (GABA), an inhibitory neurotransmitter, at GABA-B receptors in the spinal cord. This action leads to hyperpolarization of afferent terminals and presynaptic inhibition of both monosynaptic and polysynaptic reflexes, thereby reducing the frequency and severity of muscle spasms, clonus, and associated pain. Baclofen helps improve muscle function and mobility, enhancing the patient's ability to perform daily activities. Dosage must be carefully titrated to achieve optimal therapeutic effects while minimizing side effects, particularly central nervous system depression.

Uses

  • Management of spasticity in multiple sclerosis.
  • Relief of muscle spasms from spinal cord injuries.
  • Treatment of spasticity in cerebral palsy.
  • Alleviation of severe chronic spasticity of spinal origin.
