About

Bortezomib is a potent, reversible proteasome inhibitor that plays a crucial role in the treatment of certain hematologic malignancies. It works by inhibiting the 26S proteasome, a multi-catalytic proteinase complex responsible for degrading ubiquitinated proteins. This inhibition disrupts various cellular processes, including cell cycle progression, apoptosis, and angiogenesis, leading to the accumulation of misfolded proteins and ultimately inducing programmed cell death in cancer cells. Its mechanism of action makes it particularly effective against plasma cell disorders like multiple myeloma and certain lymphomas.

Clinically, bortezomib is administered intravenously or subcutaneously. Its efficacy has been demonstrated in both newly diagnosed and relapsed/refractory settings, often in combination with other chemotherapeutic agents or corticosteroids. The drug's targeted action helps to selectively eliminate malignant cells while minimizing damage to healthy tissues, although systemic side effects can occur due to the ubiquitous nature of proteasome activity. Regular monitoring of patient response and adverse events is essential throughout the treatment course.
