About

Docetaxel is an antineoplastic agent belonging to the taxoid class, primarily used in chemotherapy. It works by disrupting the microtubule network in cells, which is essential for cell division and other vital cellular functions. By stabilizing microtubules and preventing their depolymerization, docetaxel inhibits mitosis and interphase cellular functions, leading to cell cycle arrest and apoptosis in rapidly dividing cancer cells. This mechanism makes it effective against various solid tumors.

Docetaxel is administered intravenously and is metabolized primarily in the liver. Its cytotoxic action is dose-dependent and time-dependent, making careful dosing and monitoring crucial. It is often used alone or in combination with other chemotherapeutic agents or radiation therapy, depending on the specific cancer type and stage. Its efficacy has been demonstrated in clinical trials across multiple indications, improving progression-free survival and overall survival in many patients.

  • Management of non-small cell lung cancer.
  • Therapy for prostate cancer.

Safety Measures

  • Alcohol - Avoid alcohol consumption as it may exacerbate liver toxicity and other side effects associated with docetaxel.
  • Pregnancy - Docetaxel is contraindicated in pregnancy due to its genotoxic and teratogenic potential. Effective contraception is required for both male and female patients during treatment and for a period thereafter.
  • Breastfeeding - Breastfeeding is not recommended during docetaxel treatment and for a period after the last dose, as the drug may be excreted in breast milk and harm the infant.
  • Liver - Use with caution in patients with hepatic impairment; dose adjustments may be necessary due to altered metabolism and increased risk of toxicity.
  • Kidney - Use with caution in patients with severe renal impairment; close monitoring for toxicity is advised, though renal excretion is not the primary elimination route.
  • Lung - Monitor for pulmonary toxicity, including interstitial lung disease, especially in patients with pre-existing lung conditions or those receiving concomitant radiation therapy.
