About

Exemestane is a potent and irreversible steroidal aromatase inhibitor, specifically utilized in the endocrine therapy of hormone receptor-positive breast cancer in postmenopausal women. In postmenopausal women, the primary source of estrogen is the conversion of anrogens into estrogens by the aromatase enzyme, predominantly in peripheral tissues such as fat, muscle, and the liver. Exemestane works by forming a permanent covalent bond with the active site of the aromatase enzyme, thereby irreversibly inactivating it. This "suicide inhibition" leads to a significant and sustained reduction in circulating estrogen levels, effectively depriving estrogen-dependent breast cancer cells of the growth stimulus they require. By lowering estrogen, exemestane helps to inhibit tumor growth and reduce the risk of recurrence, making it a crucial agent in both adjuvant settings following tamoxifen therapy and for advanced disease.

Uses

  • Adjuvant treatment of early hormone receptor-positive breast cancer in postmenopausal women.
  • Treatment of advanced hormone receptor-positive breast cancer in postmenopausal women.
