About

Imatinib, the active component of Knilonat, is a potent and selective tyrosine kinase inhibitor, widely recognized for its efficacy in treating various hematologic malignancies and solid tumors. Its primary mechanism involves inhibiting the Bcr-Abl tyrosine kinase, a constitutively active enzyme central to the pathogenesis of chronic myeloid leukemia (CML). Additionally, it targets other receptor tyrosine kinases such as KIT and PDGFRA, making it effective against gastrointestinal stromal tumors (GIST) and certain other rare cancers.

By blocking these critical signaling pathways, imatinib prevents uncontrolled cell growth and proliferation, leading to apoptosis in cancer cells. This targeted approach has dramatically improved patient outcomes, particularly in CML, where it has transformed the disease from a rapidly fatal condition into a chronic, manageable illness for many individuals. Its specificity minimizes damage to healthy cells compared to traditional chemotherapy.

Uses

  • Treatment of chronic myeloid leukemia (CML) in various phases.
  • Management of unresectable and/or metastatic gastrointestinal stromal tumors (GIST).
  • Treatment of Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL).
  • Management of hypereosinophilic syndrome (HES) and/or chronic eosinophilic leukemia (CEL).

Directions For Use

Take this medication orally with a meal and a large glass of water to reduce gastrointestinal upset. Adhere strictly to your doctor's dosing schedule.

Benefits

  • Induces high rates of hematologic and cytogenetic remission in CML.
  • Extends progression-free and overall survival in GIST.
  • Offers a targeted therapy with improved specificity.
  • Reduces the need for more aggressive treatments in many cases.
  • Improves quality of life for patients with chronic conditions.
  • Oral administration provides convenience for long-term therapy.

Safety Measures

  • Alcohol - It is advisable to limit or avoid alcohol intake during imatinib therapy due to the potential for increased hepatotoxicity.
  • Pregnancy - Imatinib is contraindicated during pregnancy due to significant risks of fetal malformations. Women of childbearing potential must use highly effective contraception.
  • Breastfeeding - Breastfeeding is not recommended while taking imatinib and for a period after treatment cessation, as the drug can pass into breast milk.
  • Liver - Patients with pre-existing liver impairment require careful monitoring and potential dose adjustments. Regular liver function tests are crucial.
  • Kidney - Dose adjustments are generally not required for mild to moderate renal impairment, but close monitoring is recommended for severe cases.
  • Lung - Patients should be monitored for new or worsening pulmonary symptoms, as rare but serious lung toxicities like pleural effusion can occur.
