About

Carfilzomib is a proteasome inhibitor indicated for the treatment of multiple myeloma. It works by selectively and irreversibly binding to the N-terminal threonine-containing active sites of the 20S proteasome, a multi-catalytic proteinase complex responsible for degrading ubiquitinated proteins. Inhibition of the proteasome leads to an accumulation of polyubiquitinated proteins within the cell, which can trigger apoptosis, particularly in malignant plasma cells. This mechanism disrupts cellular homeostasis, leading to cell cycle arrest and programmed cell death in cancer cells, while sparing healthy cells to a greater extent. Carfilzomib is often used in patients who have received prior therapies, either as monotherapy or in combination with other anti-myeloma agents.
