About

Leuprolide is a synthetic analogue of gonadotropin-releasing hormone (GnRH), functioning as a potent GnRH receptor agonist. Its therapeutic effect stems from a biphasic action: an initial transient stimulation of pituitary GnRH receptors, followed by a sustained desensitization and downregulation of these receptors. This desensitization leads to a profound and continuous suppression of pituitary gonadotropin release, specifically luteinizing hormone (LH) and follicle-stimulating hormone (FSH). Consequently, there is a significant reduction in the production of gonadal steroids, such as testosterone in males and estrogen in females. This pharmacological castration effect is highly valuable in managing various hormone-sensitive conditions, including advanced prostate cancer, endometriosis, uterine fibroids, and central precocious puberty, by effectively lowering sex hormone levels to therapeutic targets.
