Lupniza Depot Injection contains leuprolide acetate, a synthetic analog of gonadotropin-releasing hormone (GnRH). Its mechanism of action involves an initial transient stimulation of GnRH receptors in the pituitary gland, leading to a brief surge in gonadotropin release. However, with continuous administration, it causes a profound and sustained desensitization and downregulation of these receptors. This results in a significant suppression of pituitary gonadotropin secretion, specifically luteinizing hormone (LH) and follicle-stimulating hormone (FSH).

The sustained reduction in LH and FSH levels subsequently leads to a marked decrease in the production of gonadal steroids, such as testosterone in males and estrogen in females, effectively achieving a medical castration state.
