About

Tucatinib is an oral tyrosine kinase inhibitor specifically designed to target human epidermal growth factor receptor 2 (HER2). It selectively inhibits HER2 and, to a lesser extent, epidermal growth factor receptor (EGFR) phosphorylation, thereby blocking downstream signaling pathways that promote cell growth and survival in HER2-positive cancers. Unlike some other HER2 inhibitors, tucatinib demonstrates high selectivity for HER2, which contributes to its efficacy and a more favorable safety profile. Its ability to cross the blood-brain barrier is particularly significant, making it an important treatment option for patients with HER2-positive metastatic breast cancer, especially those with brain metastases.

This medication is typically used in combination with trastuzumab and capecitabine for the treatment of adult patients with advanced unresectable or metastatic HER2-positive breast cancer, including patients with brain metastases, who have received one or more prior anti-HER2-based regimens in the metastatic setting. It offers a targeted approach to therapy, improving progression-free survival and overall survival in this challenging patient population.

Uses

  • Treatment of HER2-positive metastatic breast cancer.
  • Used in combination with trastuzumab and capecitabine.
  • Effective for patients with brain metastases.
  • For patients who have received prior anti-HER2 therapies.
