About
Tucatinib is a highly selective tyrosine kinase inhibitor (TKI) specifically designed to target the human epidermal growth factor receptor 2 (HER2) and HER3 pathways. It is primarily used in combination with other agents for the treatment of advanced HER2-positive breast cancer, particularly in patients who have previously received multiple anti-HER2 therapies. Tucatinib works by reversibly binding to the intracellular kinase domain of HER2, thereby inhibiting HER2 phosphorylation and downstream signaling pathways that drive cancer cell growth and survival.
Its selectivity for HER2 minimizes off-target effects, and its ability to cross the blood-brain barrier makes it particularly valuable for treating brain metastases, a common complication in HER2-positive breast cancer.
