About

Gemcitabine is a nucleoside analog chemotherapy agent widely used in the treatment of various solid tumors. It is indicated for pancreatic cancer, non-small cell lung cancer, breast cancer, and ovarian cancer. Gemcitabine acts as an antimetabolite, interfering with DNA synthesis and repair, ultimately leading to cell death. Once inside the cell, gemcitabine is phosphorylated to its active diphosphate and triphosphate forms. Gemcitabine diphosphate inhibits ribonucleotide reductase, an enzyme essential for DNA synthesis, thereby reducing the concentration of deoxynucleotides. Gemcitabine triphosphate is incorporated into DNA strands, leading to chain termination and inhibition of DNA polymerase. This dual mechanism of action makes gemcitabine a potent cytotoxic agent, particularly effective against rapidly dividing cancer cells. It is administered intravenously, often as part of a combination chemotherapy regimen.
