About

Gemcitabine is a nucleoside analog antimetabolite widely used in chemotherapy for various solid tumors. It acts by inhibiting DNA synthesis and repair, leading to cell death. Once inside the cell, gemcitabine is phosphorylated to its active diphosphate (dFdCDP) and triphosphate (dFdCTP) forms. The diphosphate inhibits ribonucleotide reductase, reducing the deoxyribonucleotide pool necessary for DNA synthesis. The triphosphate is incorporated into DNA, causing chain termination and inhibiting DNA polymerase, thereby disrupting DNA replication and repair mechanisms. This dual action makes gemcitabine a potent cytotoxic agent.

Its broad spectrum of activity makes it a cornerstone in the treatment of several aggressive cancers, often used alone or in combination with other chemotherapeutic agents. The drug is administered intravenously, and its efficacy is well-documented in improving patient outcomes in various advanced malignancies. Due to its mechanism of action, careful monitoring for hematological toxicities, particularly myelosuppression, is crucial during treatment.
