About

Cisplatin is a highly effective platinum-based antineoplastic agent, a cornerstone in the chemotherapy regimens for a variety of solid tumors. Its mechanism of action involves the formation of DNA adducts, leading to the creation of inter- and intra-strand crosslinks within the DNA helix. These crosslinks severely disrupt critical cellular processes such as DNA replication and transcription, ultimately triggering programmed cell death (apoptosis) in rapidly proliferating cancer cells. As a cell-cycle non-specific agent, cisplatin is effective against cells in all phases of the cell cycle, contributing to its broad spectrum of activity against malignancies like testicular, ovarian, bladder, and head and neck cancers. However, its clinical utility is balanced by a significant toxicity profile, notably severe nephrotoxicity, ototoxicity, and profound nausea and vomiting, which necessitate meticulous patient monitoring, aggressive hydration, and antiemetic prophylaxis during treatment.
