About
Pregabalin is an alpha-2-delta ligand, primarily used as an anticonvulsant and for the management of neuropathic pain. Its mechanism of action involves binding to the alpha-2-delta subunit of voltage-gated calcium channels in the central nervous system. This binding modulates the release of several excitatory neurotransmitters, including glutamate, noradrenaline, and substance P, thereby reducing neuronal hyperexcitability. This reduction in neurotransmitter release contributes to its analgesic, anxiolytic, and anticonvulsant effects. While structurally similar to GABA, pregabalin does not directly interact with GABA-A or GABA-B receptors. It is effective in treating various forms of neuropathic pain, including diabetic peripheral neuropathy and postherpetic neuralgia, and is also indicated as an adjunctive therapy for partial-onset seizures in adults. Furthermore, it is approved for the management of fibromyalgia and generalized anxiety disorder, offering a broad spectrum of therapeutic applications by stabilizing overactive nerve signals.

- Pregnancy - Use with caution; potential risks to the fetus exist. Consult a doctor to weigh benefits against risks.
- Breastfeeding - Not recommended as pregabalin is excreted in breast milk and may cause sedation in infants.
- Liver - Generally safe, but caution is advised in severe hepatic impairment. Dose adjustment is typically not required.
- Kidney - Requires dose adjustment in patients with impaired renal function due to renal excretion.
- Lung - Use with caution in patients with respiratory depression, especially when co-administered with other CNS depressants.
