About
Bortezomib is a first-in-class proteasome inhibitor, a targeted therapy crucial for the treatment of multiple myeloma and mantle cell lymphoma. It works by reversibly inhibiting the 26S proteasome, a multi-protein complex responsible for degrading ubiquitinated proteins within cells. By blocking the proteasome, bortezomib causes an accumulation of misfolded and unwanted proteins, leading to endoplasmic reticulum stress, activation of pro-apoptotic pathways, and ultimately, programmed cell death in cancer cells. This selective action against malignant cells, while sparing normal cells to a greater extent, makes it a highly effective agent, often used in combination with other drugs to improve patient outcomes and achieve deeper responses.
