About
Sitagliptin is an oral antidiabetic drug from the dipeptidyl peptidase-4 (DPP-4) inhibitor class, indicated for the treatment of type 2 diabetes mellitus. It works by selectively inhibiting the enzyme DPP-4, which normally breaks down the incretin hormones, glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). By preventing the rapid inactivation of these incretins, sitagliptin increases their levels, leading to enhanced glucose-dependent insulin release from pancreatic beta cells and reduced glucagon secretion from pancreatic alpha cells.
This mechanism helps to lower both fasting and post-meal blood glucose concentrations, improving overall glycemic control. Sitagliptin is often used as an adjunct to diet and exercise, either as monotherapy or in combination with other antidiabetic agents.
