About

Dasatinib is a potent, multi-targeted tyrosine kinase inhibitor (TKI) primarily used in the treatment of chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). It works by inhibiting the activity of several kinases, including BCR-ABL, SRC family kinases, c-KIT, EPHA2, and PDGFR-β. This inhibition prevents the proliferation of cancer cells that rely on these kinases for growth and survival. Its mechanism of action makes it effective against both imatinib-sensitive and imatinib-resistant forms of CML, offering a crucial therapeutic option for patients who have failed prior therapies or are newly diagnosed.

The drug is designed to bind to both active and inactive conformations of the BCR-ABL kinase, providing a broader inhibitory profile. This dual binding capability contributes to its efficacy in overcoming resistance mutations. Dasatinib is administered orally and is rapidly absorbed. Its targeted action helps to reduce the burden of malignant cells, leading to hematologic and cytogenetic responses in patients with these specific leukemias, thereby improving disease control and patient outcomes.

Uses

  • Treatment of newly diagnosed Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML).
  • Treatment of Ph+ CML in chronic, accelerated, or myeloid/lymphoid blast phase with resistance or intolerance to prior therapy.
  • Treatment of Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) with resistance or intolerance to prior therapy.
  • Treatment of newly diagnosed Ph+ ALL.
