About

Letrozole is a highly selective, non-steroidal aromatase inhibitor primarily used in the treatment of hormone receptor-positive breast cancer in postmenopausal women. Its mechanism of action involves competitively binding to the heme of the cytochrome P450 subunit of the aromatase enzyme, thereby inhibiting the conversion of androgens (like androstenedione and testosterone) into estrogens (estrone and estradiol). This significant reduction in circulating estrogen levels deprives estrogen-dependent breast cancer cells of the growth stimulus, leading to tumor regression or prevention of recurrence. Letrozole is a potent agent, capable of achieving greater than 90% suppression of estrogen synthesis. It is administered orally and is a cornerstone in both adjuvant and metastatic settings for estrogen receptor-positive breast cancer, demonstrating superior efficacy compared to tamoxifen in certain patient populations.

Uses

  • Adjuvant treatment of early breast cancer
