About

Palbociclib is an oral cyclin-dependent kinase (CDK) 4 and 6 inhibitor indicated for the treatment of hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative advanced or metastatic breast cancer. It works by selectively inhibiting CDK4 and CDK6, which are key regulators of cell cycle progression. This inhibition prevents cancer cells from dividing and growing, thereby slowing tumor progression. It is typically used in combination with an aromatase inhibitor or fulvestrant.

This targeted therapy helps to overcome resistance to endocrine therapy by restoring cell cycle control. Clinical studies have demonstrated that adding palbociclib to endocrine therapy significantly prolongs progression-free survival in eligible patients. Its mechanism of action focuses on disrupting the proliferation of cancer cells that rely on CDK4/6 activity for growth, offering a crucial treatment option for this specific subtype of breast cancer.

Uses

  • Treatment of HR-positive, HER2-negative advanced breast cancer.
  • Used in combination with an aromatase inhibitor as initial endocrine-based therapy.
  • Used in combination with fulvestrant after disease progression on endocrine therapy.
  • To delay disease progression in metastatic breast cancer.

Directions For Use
