About
Pregabalin is an anticonvulsant and neuropathic pain agent that is structurally analogous to the neurotransmitter gamma-aminobutyric acid (GABA). Despite this structural similarity, its mechanism of action does not involve direct binding to GABA-A or GABA-B receptors, nor does it influence GABA uptake or degradation. Instead, pregabalin primarily exerts its therapeutic effects by binding with high affinity to the alpha-2-delta subunit of voltage-gated calcium channels located in the central nervous system. This binding modulates calcium influx into presynaptic nerve terminals, consequently reducing the release of several key excitatory neurotransmitters, such as glutamate, substance P, and norepinephrine. This modulation of neurotransmitter release helps to dampen overactive nerve signals, making pregabalin highly effective in treating various forms of neuropathic pain, fibromyalgia, and as an adjunctive therapy for partial-onset seizures. The sustained-release (SR) formulation ensures a prolonged therapeutic effect, allowing for convenient once-daily dosing.
