About
Lenalidomide is an immunomodulatory drug (IMiD) with anti-neoplastic, anti-angiogenic, and immunomodulatory properties. It is primarily indicated for the treatment of multiple myeloma, myelodysplastic syndromes (MDS) with a specific chromosomal deletion (5q deletion), and mantle cell lymphoma. In multiple myeloma, lenalidomide works by directly inhibiting tumor cell proliferation, inducing apoptosis, and enhancing the activity of natural killer (NK) cells and T-cells, thereby boosting the host's anti-tumor immune response. For MDS, it selectively targets and eliminates cells with the 5q deletion, leading to erythroid responses and transfusion independence. Due to its significant teratogenic risk, lenalidomide is subject to strict risk evaluation and mitigation strategies (REMS) programs to ensure safe use.
