About

Letrozole is a potent and highly selective non-steroidal aromatase inhibitor. It works by competitively binding to the heme of the cytochrome P450 subunit of aromatase, an enzyme primarily responsible for converting androgens into estrogens in peripheral tissues. By inhibiting aromatase, letrozole significantly reduces circulating estrogen levels in postmenopausal women, thereby depriving estrogen-dependent breast cancer cells of the growth stimulus they require. This mechanism makes it a highly effective treatment for hormone receptor-positive breast cancer, particularly in the adjuvant setting after surgery, as extended adjuvant therapy, or for advanced disease. Its efficacy stems from its ability to achieve profound estrogen suppression, which is crucial for controlling the proliferation of estrogen-sensitive tumors.

Uses

  • Adjuvant treatment of hormone receptor-positive early breast cancer in postmenopausal women.
  • Extended adjuvant treatment of hormone receptor-positive early breast cancer in postmenopausal women who have received prior standard adjuvant tamoxifen therapy.
  • First-line treatment of hormone receptor-positive or unknown advanced breast cancer in postmenopausal women.
  • Treatment of advanced breast cancer in postmenopausal women with disease progression following antiestrogen therapy.
