About
Linaclotide is a guanylate cyclase-C (GC-C) agonist indicated for the treatment of chronic idiopathic constipation (CIC) and irritable bowel syndrome with constipation (IBS-C) in adults. It works by binding to the GC-C receptor on the luminal surface of intestinal epithelial cells, leading to an increase in intracellular and extracellular cyclic guanosine monophosphate (cGMP). This elevation in cGMP stimulates the secretion of chloride and bicarbonate into the intestinal lumen, increasing intestinal fluid and accelerating transit. Additionally, linaclotide has been shown to reduce visceral pain by decreasing the activity of pain-sensing nerves in the gut.
Its action is primarily local within the gastrointestinal tract, with minimal systemic absorption, contributing to a favorable safety profile. This targeted mechanism helps to alleviate constipation and associated abdominal symptoms like bloating and discomfort, improving overall bowel function and quality of life for patients suffering from these chronic conditions.
