Oxaliplatin is a third-generation platinum-based antineoplastic agent widely used in the treatment of various cancers, particularly colorectal cancer. Its mechanism of action involves the formation of DNA adducts, primarily inter- and intra-strand cross-links, which inhibit DNA replication and transcription. This leads to DNA damage, cell cycle arrest, and ultimately apoptosis in rapidly proliferating cancer cells. Unlike cisplatin and carboplatin, oxaliplatin has a distinct spectrum of activity and a different toxicity profile, notably less nephrotoxicity and ototoxicity.
