About

Temozolomide is an oral alkylating agent used in the treatment of certain brain tumors, particularly glioblastoma multiforme and anaplastic astrocytoma. It is a prodrug that rapidly converts to its active metabolite, MTIC (5-(3-methyl-1-triazen-1-yl)imidazole-4-carboxamide), at physiological pH. MTIC then methylates DNA at various sites, primarily at the O6 and N7 positions of guanine. This methylation leads to DNA damage and subsequent activation of DNA repair pathways. If the repair mechanisms are overwhelmed or faulty, the DNA damage triggers apoptosis (programmed cell death) in tumor cells. Temozolomide's ability to cross the blood-brain barrier effectively makes it a valuable therapeutic option for primary and recurrent brain malignancies, often used in combination with radiation therapy.

Directions For Use
