About

Thalidomide is an immunomodulatory drug with anti-inflammatory and anti-angiogenic properties. Initially infamous for its teratogenic effects, it has been repurposed and is now used under strict regulatory programs for specific conditions. Its primary indications include the treatment of multiple myeloma, often in combination with dexamethasone, and for the acute treatment of cutaneous manifestations of moderate to severe erythema nodosum leprosum (ENL). The exact mechanisms by which thalidomide exerts its therapeutic effects are complex and involve modulation of cytokine production, inhibition of angiogenesis, and direct effects on immune cells. Due to its severe teratogenicity, a comprehensive risk management program is mandatory for its prescription and dispensing to prevent fetal exposure. This program ensures that only patients who meet specific criteria and adhere to strict contraceptive measures receive the medication.
