About

Deferasirox is an orally active iron chelator specifically designed to treat chronic iron overload resulting from frequent blood transfusions, a common complication in patients with conditions such as thalassemia, sickle cell disease, and myelodysplastic syndromes. Its mechanism involves forming a stable, high-affinity complex with excess iron in the body, which is then primarily excreted via the feces. This chelation process is vital for preventing and mitigating iron-induced organ damage, particularly to the heart, liver, and endocrine glands, which can lead to severe morbidity and mortality if left untreated. The convenience of its oral administration significantly enhances patient adherence compared to traditional parenteral chelators, making it a cornerstone therapy for long-term iron management and improving the overall prognosis and quality of life for affected individuals.

Uses

  • Chronic iron overload due to blood transfusions (transfusional hemosiderosis)
  • Non-transfusion-dependent thalassemia syndromes with iron overload
  • To prevent iron-related organ damage
  • Management of iron accumulation in various hematological disorders

Directions For Use

Take orally once daily on an empty stomach, at least 30 minutes before food, at the same time each day. Disperse in water or juice.

Benefits

  • Effectively removes excess iron
  • Prevents iron-induced organ damage
  • Oral administration for convenience
  • Improves quality of life for patients
  • Reduces risk of cardiac complications
  • Maintains iron balance

Side Effects

  • Nausea
  • Vomiting
  • Diarrhea
  • Abdominal pain
  • Rash
  • Increased serum creatinine (kidney impairment)
  • Elevated liver enzymes (liver impairment)
  • Hearing loss
  • Vision disturbances
  • Proteinuria
  • Headache
  • Fatigue

Safety Measures

  • Alcohol - Avoid or limit alcohol consumption as it can increase the risk of liver toxicity, which is a known side effect of deferasirox.
  • Pregnancy - Not recommended during pregnancy unless the potential benefit justifies the potential risk to the fetus. Use effective contraception.
  • Breastfeeding - Not recommended during breastfeeding as it is unknown if deferasirox is excreted in human milk and could harm the infant.
  • Liver - Use with caution in patients with hepatic impairment. Monitor liver function tests closely; dose adjustment or discontinuation may be necessary.
  • Kidney - Contraindicated in patients with severe renal impairment (creatinine clearance <10 mL/min). Monitor serum creatinine and proteinuria regularly; dose adjustment may be needed.
  • Lung - No specific lung-related safety concerns are typically associated with deferasirox.
