About

Imatinib is a groundbreaking tyrosine kinase inhibitor that revolutionized the treatment of several cancers, most notably chronic myeloid leukemia (CML) and gastrointestinal stromal tumors (GIST). Its highly targeted action stems from its ability to selectively inhibit the Bcr-Abl tyrosine kinase, which is the oncogenic driver in CML. By blocking this abnormal protein, imatinib prevents the uncontrolled proliferation of leukemic cells and induces apoptosis, leading to deep and durable remissions.

Beyond CML, imatinib also inhibits other receptor tyrosine kinases, including KIT and PDGFRA, which are key drivers in the pathogenesis of GIST. This targeted inhibition effectively suppresses tumor growth and survival in GIST patients. The introduction of imatinib marked a paradigm shift in cancer therapy, demonstrating the power of molecularly targeted drugs to achieve significant clinical responses with a more favorable side effect profile compared to conventional chemotherapy. It is also used in other rare conditions like myelodysplastic/myeloproliferative diseases and hypereosinophilic syndrome.
