About
Asparaginase is an enzyme-based antineoplastic agent critical in the treatment of acute lymphoblastic leukemia (ALL). Its unique mechanism of action involves depleting asparagine, a non-essential amino acid that is, paradoxically, essential for the survival and proliferation of certain leukemic cells. These specific leukemic cells lack the enzyme asparagine synthetase, rendering them unable to synthesize their own asparagine and thus dependent on exogenous sources. Normal healthy cells, however, possess asparagine synthetase and are largely unaffected by asparagine depletion. By hydrolyzing asparagine into aspartic acid and ammonia, asparaginase effectively starves these leukemic cells, inhibiting their protein synthesis and ultimately leading to cell death. It is a vital component of multi-agent chemotherapy regimens for both pediatric and adult ALL, administered intravenously or intramuscularly.
