About
Imatinib is a potent tyrosine kinase inhibitor that selectively targets the Bcr-Abl fusion protein, a constitutively active tyrosine kinase central to the pathogenesis of chronic myeloid leukemia (CML) and Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL). It also inhibits other tyrosine kinases, including c-KIT and PDGF-R, making it effective in treating gastrointestinal stromal tumors (GIST) and certain myelodysplastic/myeloproliferative diseases. By blocking the ATP-binding site of these kinases, imatinib prevents the phosphorylation of downstream signaling proteins, thereby inhibiting cell proliferation and inducing apoptosis in cancer cells that rely on these pathways for growth and survival. Its introduction revolutionized the treatment of CML, significantly improving patient outcomes.
Uses
- Treatment of newly diagnosed Philadelphia chromosome-positive chronic myeloid leukemia (Ph+ CML).
- Treatment of Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL).
- Treatment of unresectable and/or metastatic malignant gastrointestinal stromal tumors (GIST).
- Treatment of myelodysplastic/myeloproliferative diseases (MDS/MPD).
Directions For Use
Take orally with a meal and a large glass of water to minimize gastrointestinal irritation. Dosage varies based on the condition being treated and patient tolerance.

Safety Measures
- Alcohol - Avoid or limit alcohol consumption as it can increase the risk of liver toxicity, a known side effect of imatinib.
- Pregnancy - Contraindicated due to potential for fetal harm. Women of childbearing potential must use effective contraception.
- Breastfeeding - Not recommended as imatinib is excreted in breast milk and could cause serious adverse effects in the infant.
- Liver - Use with caution in patients with hepatic impairment; dosage adjustments may be necessary. Monitor liver function closely.
- Kidney - Use with caution in patients with renal impairment; dosage adjustments may be required. Monitor kidney function.
- Lung - Can rarely cause interstitial lung disease or pulmonary hypertension; monitor for new or worsening respiratory symptoms.
