About

Temozolomide is an oral alkylating agent that serves as a prodrug, spontaneously converting to its active metabolite, MTIC (5-(3-methyl-1-triazen-1-yl)imidazole-4-carboxamide), at physiological pH. MTIC then methylates DNA at various sites, primarily at the O6 position of guanine, leading to DNA damage and subsequent apoptosis in rapidly dividing cells. This mechanism makes it particularly effective in the treatment of certain brain tumors, including glioblastoma multiforme and anaplastic astrocytoma, due to its ability to cross the blood-brain barrier. It is often used in combination with radiation therapy for newly diagnosed glioblastoma.
