PALBOTOR is an oral cyclin-dependent kinase (CDK) 4 and 6 inhibitor, representing a significant advancement in targeted therapy for hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced or metastatic breast cancer. This medication works by selectively inhibiting CDK4 and CDK6, key enzymes that regulate cell cycle progression. In cancer cells, these kinases are often overactive, driving uncontrolled proliferation. By blocking CDK4 and CDK6, PALBOTOR prevents the phosphorylation of retinoblastoma protein (Rb), which is essential for the cell to transition from the G1 phase to the S phase of the cell cycle. This inhibition effectively halts cell proliferation and induces cellular senescence, thereby slowing down tumor growth and progression. PALBOTOR is typically used in combination with endocrine therapy, such as an aromatase inhibitor or fulvestrant, to enhance its therapeutic effect and improve progression-free survival in eligible patients.

Directions For Use

Take orally once daily with food for 21 consecutive days, followed by 7 days off, to complete a 28-day cycle.
