About

Denosumab is a human monoclonal antibody that specifically targets and binds to RANK Ligand (RANKL), a protein essential for the formation, function, and survival of osteoclasts. By inhibiting RANKL, denosumab prevents osteoclast activation and differentiation, thereby reducing bone resorption and increasing bone mineral density. This mechanism makes it highly effective in treating postmenopausal osteoporosis, increasing bone mass in men with osteoporosis, and managing bone loss associated with hormone ablation therapy for prostate or breast cancer.

It is also used to prevent skeletal-related events (SREs) such as pathological fractures, spinal cord compression, or the need for radiation or surgery to bone, in patients with bone metastases from solid tumors. Administered subcutaneously, denosumab provides a sustained reduction in bone turnover markers, offering a significant therapeutic option for various bone-related conditions.

Uses

  • Treatment of postmenopausal osteoporosis
  • Increased bone mass in men with osteoporosis
  • Bone loss associated with hormone ablation therapy for prostate or breast cancer
  • Prevention of skeletal-related events in cancer patients with bone metastases

  • Liver - No specific dose adjustment is required for patients with liver impairment. Use with caution and monitor for any adverse effects.
  • Kidney - Use with caution in patients with severe kidney impairment (CrCl < 30 mL/min) or on dialysis, as they are at higher risk of hypocalcemia. Calcium and vitamin D supplementation is crucial.
  • Lung - No specific contraindications or dose adjustments related to lung conditions. However, general health status should be considered.
