About

Regorafenib is an oral multi-kinase inhibitor that targets various kinases involved in oncogenesis, tumor angiogenesis, and maintenance of the tumor microenvironment. It inhibits several receptor tyrosine kinases, including VEGFR1, VEGFR2, VEGFR3, KIT, RET, PDGFR-beta, and FGFR, as well as the intracellular kinase BRAF. By blocking these pathways, regorafenib disrupts tumor cell proliferation, inhibits the formation of new blood vessels that supply tumors, and modulates the tumor's surrounding environment, ultimately leading to tumor growth inhibition and cell death. Its broad spectrum of kinase inhibition makes it effective against several advanced solid tumors.

Regorafenib is indicated for the treatment of metastatic colorectal cancer (mCRC) that has progressed on prior therapies, unresectable or metastatic gastrointestinal stromal tumors (GIST) after prior treatment with imatinib and sunitinib, and hepatocellular carcinoma (HCC) in patients previously treated with sorafenib. It represents an important option for patients with refractory cancers, extending progression-free and overall survival in these challenging settings. Close monitoring for adverse events, particularly hand-foot skin reaction and hypertension, is crucial.

Directions For Use

  • Kidney - No dose adjustment is required for mild to moderate renal impairment; use with caution in severe impairment.
  • Lung - Monitor for new or worsening pulmonary symptoms, including interstitial lung disease, though rare.
