About

Temozolomide is an oral alkylating agent primarily used in the treatment of certain brain tumors. It functions by interfering with the DNA of cancer cells, preventing their growth and division, ultimately leading to cell death. This cytotoxic effect is particularly valuable in rapidly dividing cells, such as those found in aggressive gliomas. As a prodrug, it is spontaneously hydrolyzed at physiological pH to its active compound, MTIC (5-(3-methyl-1-triazen-1-yl)imidazole-4-carboxamide). Temozolomide's ability to cross the blood-brain barrier makes it an effective option for central nervous system malignancies. It is often utilized in combination with radiation therapy or as a monotherapy, depending on the specific tumor type and stage. Its mechanism targets DNA replication and transcription, making it a cornerstone in the management of glioblastoma multiforme and anaplastic astrocytoma.
