About

Temozolomide is an oral alkylating agent used in the treatment of certain aggressive brain tumors, including newly diagnosed glioblastoma multiforme and refractory anaplastic astrocytoma. It functions as a prodrug that rapidly converts to its active metabolite, MTIC (5-(3-methyl-1-triazen-1-yl)imidazole-4-carboxamide), at physiological pH within the body. MTIC exerts its potent cytotoxic effects by methylating DNA at various critical sites, primarily at the O6 and N7 positions of guanine. This specific methylation leads to DNA damage and subsequent apoptosis (programmed cell death) in rapidly dividing tumor cells, thereby inhibiting tumor growth. Temozolomide's unique ability to effectively cross the blood-brain barrier makes it particularly valuable and effective for treating central nervous system malignancies. It is often used in combination with radiation therapy for newly diagnosed glioblastoma, significantly improving patient outcomes and survival rates.
