About

Fludarabine is a purine analog antimetabolite indicated for the treatment of B-cell chronic lymphocytic leukemia (CLL) in patients who have not responded to or whose disease has progressed during treatment with at least one prior standard alkylating agent-containing regimen. It acts by interfering with DNA synthesis and repair, leading to cell death, particularly in rapidly dividing cells like cancer cells. Fludarabine is phosphorylated intracellularly to its active metabolite, fludarabine triphosphate, which inhibits DNA polymerase alpha, DNA primase, and ribonucleotide reductase, thereby disrupting DNA replication and repair.

This cytotoxic action makes it a cornerstone in the treatment of CLL, often used in combination with other agents. Its efficacy in inducing remissions and improving survival in CLL patients has been well-documented.
